USDA Rust Barcoding Project.

The Arthur Fungarium (PUR) at Purdue University is home to the world's most valuable and comprehensive collection of rust fungi. (Puccinales). The USDA Rust Barcoding Project is focused on generating DNA barcodes for rust fungi by utilizing collections contained within the Arthur Fungarium. Molecular work on rust fungi has historically proved challenging, and therefore many basic biological, taxonomic, phylogenetic, and evolutionary questions remain obscure. Using methods and primers developed by the Aime lab, barcodes for ca. 2000 species of rust fungi from across the class and from around the world will be generated. Overall, this project seeks to: 1) facilitate rapid identification of newly introduced rust diseases in the U.S., 2) clarify species limits and relationships between taxa that currently rely solely on classic morphological host-based identifications, 3) link disjunct spore types from certain species to enable a complete understanding of the species' life cycle. All barcodes generated for this project will be uploaded to NCBI GenBank and to a rust-specific database hosted by PUR. Here we present a selection of results from this project, including previously unsequenced taxa and notable systematic circumscriptions.   Keywords: DNA; barcode; collections; fungarium; taxonomy; rust fungi.
