Phylogenomics

Phylogenetic relationships of Mexican species of Cladocolea Tiegh. and Struthanthus Mart. (Loranthaceae).

Cladocolea Tiegh. is a neotropical genus of the Loranthaceae family, which includes 30 species distributed from northern Mexico to South America, of which 20 are found in Mexico. Several authors consider that Cladocolea is closely related to Struthanthus, which differ by having simple, determinate inflorescences and the absence of bracteoles, whereas Struthanthus has inflorescences composed of triads, indeterminate and with bracteoles on the lateral flowers of the triads. Despite the well-defined boundaries between the two genera, some species of Cladocolea have been described with indeterminate inflorescences. Several phylogenetic studies have studied the evolutionary relationships of the genera of Loranthaceae, where Cladocolea and Struthanthus are sister groups, however few species have been included. The goal of this research is to test the existing taxonomic circumscriptions and gain insight into the relationships of Cladocolea and Struthanthus.
Voucher specimens and leaf samples for DNA extraction were obtained from live plants in the field. Leaves were obtained for 19 of the 20 species found in Mexico, also 10 species of Struthanthus, two of Phthirusa and one of Passovia and one Oryctanthus were included. Total DNA was extracted using Qiagen DNeasy Plant Kit, samples were diluted and quantified. Nuclear DNA region (ITS) and a chloroplast region (trnL-trnF) were amplified. A data matrix was prepared with the sequences obtained and analyzed by Bayesian inference (BI) and maximum likelihood (ML) methods.
The results obtained by Maximum Likelihood analysis show that Cladocolea and Struthanthus are a monophyletic group. Five species of Cladocolea (C. cupulata, C. diversifolia, C. pringlei, C. racemosa and C. spathiflora) are nested within Struthanthus, some of these have indeterminate inflorescences. The Cladocolea-Struthanthus clade could be divided in two monophyletic groups. The first consists of Struthanthus species and Cladocolea species with scandent habit, whose branches form secondary haustorial connections derived from adventitious roots; the second group consisting of Cladocolea species with shrubby growth, with a single haustorium derived from the embryo. The results obtained by Bayesian Inference recover a monophyletic group formed by Cladocolea and Struthanthus, however the relationships between these two genera were not well resolved. In both analyses, the genera Oryctanthus, Passovia, and Phthirusa occupy a basal position. Further studies with additional species and data (morphological and anatomical) are expected to clarify the relationships between these two genera.
