Using RADseq for ex situ conservation of critically endangered cycads.

Southern Africa is the center of diversity for Encephalartos, a genus of cycads in the family Zamiaceae. Most are near extinction, making it imperative that conservation strategies are implemented to prevent the loss of these species. Individuals have been poached from their habitats to the extent that wild populations are either extirpated or may no longer support adequate levels of genetic diversity. To evaluate the diversity of extant plants, we have sampled from public and private collections in South Africa and the United States. These holdings collectively encompass the majority of the known individuals of five of the most critically endangered species. Due to the massive size of cycad genomes and the lack of a reference genome, we are using double digest RAD-sequencing to prepare libraries for Illumina sequencing. Our goals are to assess the genetic structure and diversity within each of these species and to identify clones. Our analysis will inform management of ex situ assurance colonies to protect against extinction and ensure that maximum genetic diversity is maintained.
