High-frequency shoot regeneration from leaf explants and assessment of clonal fidelity of micropropagated plantlets of lamb’s lettuce (Valerianella locusta L.).

Lamb's Lettuce (Valerianella locusta L.), an important winter-green in Europe, can provide an essential heterologous gene expression platform for functional genomics related studies. In this study, we report an efficient and reliable shoot induction and plant regeneration protocol from leaf explants of Valerianella locusta L. Leaf explants of different ages of lamb's lettuce were cultured on Murashige and Skoog's medium supplemented with Gamborg's vitamin (MSB5) and augmented with different concentrations of 2,4-D (2 - 10 μM) and Kinetin (0 - 5 μM) for callus induction. Two-week-old cotyledon leaf explants induced morphogenic callus formation in MSB5 containing 5 µM 2,4-D and 5 µM Kinetin. The fragile callus tissues were transferred to MSB5 supplemented with 2 µM BAP and 2 µM IBA for shoot induction.   The well-developed shoots were excised from the callus base, elongated, and rooted in MSB5 medium.  The genetic fidelity of the donor plants and regenerants were confirmed with Start Codon Targeted (SCOT) and Random Amplified Polymorphic DNA (RAPD) molecular markers. The simple, high-frequency, and reproducible protocol presented in this study should offer a feasible system for Agrobacterium tumefaciens mediated genetic transformation in lamb's lettuce.
