Comparative Genomics/Transcriptomics Pradhan, Bijaya [1], Nelson, Dillon [1], Neupane, Surendra [2], Nepal, Madhav [3]. Characterization of GmWRKYs involved in a 3-way Interaction of Soybean, Soybean-Aphid and Soybean-Cyst-Nematode. Soybean aphid (SBA) and Soybean-cyst-nematode (SCN) are the two major pests of soybean, affecting soybean production. One recent study has reported one WRKY gene (GmWRKY) promoting SCN resistance in soybean, however, GmWRKYs involved in a three-way interaction of soybean, SBA and SCN are yet to be charcaterized. In the present study, we aimed to determine the GmWRKYs involved in such interaction. A greenhouse water bath experiment was conducted, and pathogen interactions were examined in randomized block design. The SCN-resistant and SCN susceptible soybean cultivars were used with SBA, SCN, and both SCN and SBA. The treatments receiving SCN were infested with 2000 nematode eggs, and 15 (biotype1) SBA were infested with the treatments receiving soybean-aphids at the second trifoliate growth stage. Differentially expressed genes using the biotic stress pathway into MapMan showed that the plants treated with SBA and SCN in resistant cultivars showed the involvement of soybean WRKYs (GmWRKYs) in gene regulation. Twelve GmWRKYs were up-regulated at five days post infestation (dpi), and 14 GmWRKYs were down-regulated in 30 dpi. Altogether, we found 26 GmWRKYs involved in gene regulation in the 3-way interaction. Validation of these genes using qRT- PCR data and statistical analysis are under progress. In this paper, we will discuss the final outcomes of the qPCR data and determination of the GmWRKYs of the major effects in the three-way interaction. Log in to add this item to your schedule
