Phytochemical Ogunda, Ari [1], Liebelt, Donna [2], Waddell, Eric [3], Doherty, Colleen [1]. New Annotation, New Hopportunity: Iso-Seq Annotation of Humulus lupulus. Phytochemicals are a key contributor to the world nutraceutical supply; plant derivatives are in a variety of consumer products from food and health supplements to prescription drugs. We are interested in understanding how environmental perturbations impact the regulation and production of phytochemical pathways. However, most plant species prized for their phytochemical profiles lack the quality genome annotations needed to understand the regulatory networks controlling environmental responses. We have developed a pipeline using Iso-Seq to improve genome annotation in Humulus lupulus or hop. While known for their role in beer production, hop plants are also a rich source of phytochemical nutraceuticals, including antioxidants, antimicrobials, and essential oils. The production and accumulation of many phytochemicals are tightly controlled by the plant’s signaling and regulatory systems based on environmental conditions. Therefore, the yield and quality of these chemicals fluctuate in response to a dynamic environment. In particular, we seek to understand how changing climates affect the composition of specialized metabolites, particularly those with the potential to improve human health. We have performed Iso-Seq on five different hop tissues at two times of the day. We identified over 50,000 unique transcript reads. Over 60% of these reads could not be mapped to the genome, suggesting novel annotations. Included in this data are new candidate transcripts and isoforms. Many of these are supported by expression across multiple tissues. We compare the relative contribution of the time of day and tissue type and find that both play a significant role in improving annotation. Log in to add this item to your schedule
