Phylogenomics Musker, Seth [1]. Next-generation phylogenomics of the genus Erica (Ericaceae) in the Cape Floristic Region, South Africa. Erica (Ericaceae) is by far the largest plant genus in the hyper-diverse Cape Floristic Region, with close to 700 described species, representing roughly 80% of all Erica species. This radiation began with a single colonisation event roughly 10 mya and many diverse clades have much younger ages, rendering traditional phylogenetics incapable of clearly resolving their phylogenetic relationships (Pirie et al. 2016). To address this, I have focused on the Erica abietina-viscaria clade which shows a typical pattern of recent and rapid radiation into diverse phenotypes and environments, and for which a Sanger sequencing-derived, relatively poorly resolved phylogenetic hypothesis exists (Pirie et al. 2017). Building on the target capture work of Kadlec et al. (2017), I have expanded their 134-gene target set to include 100 genes derived from the Angiosperm K353 set (Johnson et al. 2019, McLay et al. 2020) and 59 genes mined using MarkerMiner (Chamala et al. 2015) from recently published Rhododendron genomes. These new gene sets were vetted by mapping off target reads from Kadlec et al. (2017) to determine their presence in Erica and exclude putative multi-copy genes. In parallel, I have produced a Genotyping-by-Sequencing (GBS; Elshire et al. 2011) data set, for which I will present initial results, and explore the possibilities for the use of GBS data to identify putative “speciation genes” for integration into a target capture approach.
