Tissue-Specific Metabolite Profiling of Bloodroot (Sanguinaria canadensis L., Papaveraceae) Extracts using Liquid Chromatography-Tandem Mass Spectrometry (LC/MS).

Rhizome extracts from bloodroot (Sanguinaria canadensis L.) have been used in both Native American and contemporary Western medicinal contexts. Today they are valued for their potential antibacterial and anticarcinogenic properties, attributed to benzophenanthridine alkaloids. The purposes of this investigation were: (1) to use liquid chromatography-tandem mass spectrometry (LC/MS) to profile benzophenanthridine alkaloid metabolites from extracts of flower, leaf, rhizome (stem), and basal complex; and, (2) to compare those profiles from different populations. Bloodroot was collected from six different locations around Ames, Iowa in early spring 2014. Plants were separated into four tissues (flower, leaf, basal complex, and rhizome), and alkaloids were extracted using an 8:2 ethanol:water solvent. Nine quaternary benzophenanthridine alkaloids were detected and identified (sanguinarine, magnoflorine, chelerythrine, protopine, chelirubine, sanguirubine, chelitutine, macarpine and sanguilutine). Quantification parameters were optimized using multiple reactions monitoring (MRM) to accurately quantify alkaloids from each extract. Principal component analysis was performed to visualize the variance in alkaloid distribution among the different tissue types. Alkaloid distribution patterns varied among the different tissues within the same plant, suggesting alkaloid profiling could be used as a probe to distinguish different tissues from the same plant. Metabolite profiles were markedly different between aboveground and belowground structures. With few exceptions, plants collected from different sites showed no differences in alkaloid profiles. Rhizomes contained the most abundant alkaloids.
