Recent Topics Posters Kaffer, J.P. [1], Carey, Sarah [2], Harkess, Alex [3]. The Genome of Wolffia microscopica, One of the Smallest Flowering Plant Species . Wolffia microscopica is the smallest flowering plant species and is able to clonally reproduce in as little as 12 hours. In order to explore genomic features that might underlie rapid reproduction, nearly 166X coverage of the W. microscopica genome was generated on the Illumina NovaSeq6000 using paired-end 150 nt reads. Genome size, ploidy, and heterozygosity was assessed using Jellyfish and GenomeScope. Chloroplast and mitochondrial genomes were assembled using NOVOplasty, followed by annotation using GeSeq. The estimated genome size was 508 Mb, of which 14% was repetitive content. The chloroplast genome is 167,608 base pairs long, with 37 genes, similar to other Wolffia species. The de novo assembly of the genome was one using Meraculous which found the genome to be 592MB and the main contig was N50 1.5Kb and L50 111851Kb. In the future, these data will guide a chromosome-scale genome assembly for W. microscopica as a framework for the discovery of genes underlying these unique biological traits.
