Recent Topics Posters Martinez-Sagarra, Gloria [1], Devesa, Juan A. [1]. Unexpectedly high cytogenetic variation in Festuca L. (Poaceae) using flow cytometry and chromosome count. Polyploidy, or whole-genome duplication (WGD), has long been recognised as a major force in angiosperm evolution. This phenomenon has played an important evolutionary role in the genus Festuca L. (Poaceae), in which several ploidy levels (ranging from 2n = 2x = 14 to 2n = 12x = 84) have been detected to date. We studied the genome size and ploidy level of two morphologically complex subspecies belonging to the Festuca yvesii polyploid group by flow cytometry (FCM) and chromosome counting. Our results reveal an unexpectly high cytogenetic diversity among populations for each taxon. The genome size estimates varied from 2C = 13.60 to 31.91 pg, and five ploidy levels were identified (from 6x to 14x). The 2C values were positively correlationated with chromosome number counts shown herein, in line with other previous studies. We report the highest ploidy level (2n = 14x = 98) found for the genus Festuca and previously unknown cytotypes are also reported for both species. This study opens up the possibility of further research with which to explore cytogenetic diversity in fescue species with complex morphological pattern variation, by considering the advantages of the flow cytometric method in the ploidy check at the population level.
