A novel natural product reservoir: secondary metabolite extracts from a zygomycete, Basidiobolus meristosporus, show selective antibacterial activity. Investigations into fungal-derived secondary metabolites (SMs) have yielded important biologically active compounds that are currently used in medicine. Fungal SM production has been investigated primarily in Dikarya, but genomic analyses of Basidiobolus meristosporus (Zoopagomycota) indicate enrichment of biosynthetic gene clusters linked to SM production. Here we test SM extracts from B. meristosporus against a panel of World Health Organization priority yeast and bacterial pathogens. B. meristosporus was grow in five different media conditions, including Yeast Powder Soluble Starch Broth (YPSS), Yeast Malt Broth, Malt Extract Broth, Potato Dextrose Broth, and Cornmeal Broth. Chemical extracts from tissue and culture filtrate were analyzed via liquid chromatography/tandem mass spectrometry (LC-MSMS) to assess differential production of SMs in response to media composition. Chemical extracts from mycelial tissue across all media conditions were then screened for bioactivity using microtiter plate assays. Assays were selected for identification of extracts with antibacterial, antifungal, cytotoxic, or immunomodulating activities. Positive hits were detected during antibacterial growth inhibition assays, especially in samples from YPSS conditions, and bioactive crude extracts were fractionated via high performance liquid chromatography (HPLC) for further screening. LC-MSMS profiles and growth inhibition assays indicate distinct SM profiles and bioactivity of extracts between growth conditions. Log in to add this item to your schedule
